Introduction

The MultiMACS Cell24 Separator Plus is a semi-automated magnetic cell separator that enables magnetic cell
isolations based on MACS MicroBeads and MACS Columns. It is the first step to automated cell separation and
allows for parallel multisample magnetic cell isolations using MACS Technology. The instrument contains a
permanent 24-well magnet that induces a high-gradient magnetic field within columns. The field is strong
enough to retain cells labeled with MACS MicroBeads in the columns. After transfer of columns to the MACS
Elution Chamber of the MACS Elution Station, magnetically labeled cells can be eluted from columns via a small
vacuum impulse.
The instrument is designed for isolation of virtually any cell type from any source, giving reliable and
standardized results. It is compatible with many downstream applications such as molecular and functional
assays, chimerism analysis, and flow cytometry. Positive or untouched isolations can be performed using MACS
Cell Separation Reagents, including StraightFrom MicroBeads. Thereby, cells can be isolated from multiple
sources: dissociated tissues, single cell-suspensions, peripheral blood mononuclear cells (PBMCs), or directly
from blood products including whole blood, buffy coat, LRSC, and Leukopak®. Separations can be performed
with the Multi-24 Column Block for simultaneous isolation of up to 24 samples or by splitting one large volume
sample. Alternatively, up to 12 LS, LD, or Whole Blood Columns can be used in combination with the
MultiMACS Single-Column Adapter. Deep well plates and 5 mL tubes can be used for sample collection.
